statins: newer roles including lipid lowering therapy
نویسندگان
چکیده
statins are most effective pharmacologically agents to decrease total plasma cholesterol level by competitive inhibition of hmg-co a reductase. apart from the well-known ldl and cholesterol lowering effect, statins have been postulated to exert other beneficial effects also called pleotropic or non-lipid effects. better understanding of various pleotropic effects of statins has prompted a new surge of interest in their use to treat or prevent a wide range of chronic and life threatening disorders. their effectiveness in treating these disorders suggests that the benefits of statins may not be limited to cholesterol lowering. however at present although the clinical implication of this beneficial non-lipid effect seems promising yet properly designed, large multicentre, prospective, control trials are going on to validate the use of statins for indication other than primary and secondary prevention of vascular disease.

متن کاملBenefits of statins beyond lipid lowering
Increasing evidence has accumulated suggesting that inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase, or statins, have therapeutic ‘pleiotropic’ effects independent of cholesterol lowering. These include anti-inflammatory and antioxidative properties, improvement of endothelial function and increased nitric oxide bioavailability. In addition to elucidating underlying mech...
متن کاملStatins and cardiovascular diseases: the multiple effects of lipid-lowering therapy by statins.
Cholesterol lowering involving different therapies improves the clinical outcome of patients. To define the underlying pathomechanism, we studied whether treatment with statins was associated with changes in blood thrombogenicity, endothelial dysfunction and soluble adhesion molecule levels. Fifty hypercholesterolemic patients were treated with pravastatin (40 mg/day, n=24) or simvastatin (20 m...
متن کاملLipid lowering: statins and the future.
Cardiovascular disease (CVD) is the leading cause of death in developed and many developing countries. Between 1970 and 1992 there has been a slow decline in some areas, notably North America, Western Europe, Australia, and New Zealand. However, CVD mortality has recently increased in Eastern and Central Europe. A whole spectrum of therapeutic tools is available to manage CVD including thrombol...
متن کاملLipid-lowering therapy with statins, a new approach to antiarrhythmic therapy.
Inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase (statins) are the most effective and best-tolerated drugs to treat elevated levels of low-density lipoprotein cholesterol (LDL-C). In addition, they exhibit other effects unrelated to their lipid lowering effects (pleiotropic actions). In recent years, experimental and clinical evidence demonstrates that statins exert antia...
متن کاملBeyond lipid lowering: pleiotropic effects of statins in heart failure
In the 1980s, statins were introduced in clinical practice as lipidloweringmedication. Since then, several additional, pleiotropic, effects of statins have been described, including angiogenic, antiarrhythmic, antibacterial, anti-inflammatory, antimitotic, antioxidative, antithrombotic, CRP-lowering, immunomodulatory and vascular protective (stabilisation of the atheroma plaque) activity, inhib...
